Sumario:The Oocyte have the inherent ability to reprogram the sperm nucleus to form an embryo, which result in a new organism; nevertheless, also it has the ability to reprogram the nucleus of differentiated somatic cell, as it has been shown in Somatic Cell Nuclear Transfer (SCNT). Cytoplasmic extracts of Germinal Vesicle oocytes (VG) and Metaphase II (MII) oocytes have been used for reprogramming of cumulus cells or fibroblasts, they used as karyoplasts for production cloning embryos of mammalian, improving the growth rate and embryos quality. In this study, the effect fibroblast of Ovis aries exposure to homologues MII oocytes extracts, used as karioplasts for in vitro production of embryos by handmade cloning, The stages reached of embryos development was 3 to 8, 10 to 12 cells and morula. No significant difference (P>0.05) between groups with respect to the embryos development stage; however, to analyze the relationship between the number of nucleus and the number of blastomere of embryos, which showed cytoplasmic fragmentation; It was observed that cloned embryos from fibroblasts treated with MII oocytes extracts (0.58) showed better ratio compared to those untreated (0.45) and partenogenetic. Exposure of fibroblasts to extracts MII oocytes, improved communication between the nucleus and cytoplasm from cloning embryos of Ovis aries; which was reflected in a higher proportion of blastomeres with nucleus in the fragmentation embryos. It is noteworthy that depending on the magnitude of cytoplasmic fragmentation may or may not compromise the viability of the embryo development.
