description The phylum Cnidarian contains about 11,000 species, the jellyfishes has cells specialized (cnidocitos) that contain toxins; this toxins have the potential to be a tool with which to study some complex physiological systems, in addition to being used for the manufacture of new drugs and obtaining knowledge of its operation helps protect subsequent poisonings on humans; the main objective of this study was to evaluate and characterize toxins from the venom of the jellyfish Chrysaora sp. found in the lagoon Mandinga, Veracruz, Mexico; to achieve this objective, the study of biological aspects was necessary, such as physic-chemical and morphological parameters, also histological analysis of the jellyfishes collected in the field, the species was determined by analyzing the mitochondrial subunit cytochrome c oxidase III (COIII) molecule and toxicity was evaluated in vivo tests, using the Danio rerio fish as a biological model, and in vitro assays measuring the hemolytic and phospholipase A2 activities. Proteins present in the venom were analyzed by electrophoresis in one and two dimensions, obtaining the peptide fingerprint and sequencing tryptic peptide by De Novo sequencing. The conditions suitable for aquarium of this jellyfish, is a volume greater than 545L, a salinity of 28°/ºº and a temperature of 30°C. The main affectation was the bell Evert of the hood observed during acclimatization. Six color patterns were recognized that could be related to salinity, also the size of the jellyfish varies according to the period of collection, the size was a larger (8.962.4 cm) during the rainy season and the females having a predominance with gonads mature state, the specie of the jellyfish in this coastal lagoon is Chrysaora sp.; its venom causes convulsions, paralysis, bleeding in the eyes and disturbances in circulation on the Danio rerio fish, mainly affects the gill tissue, cause hyperemia in pseudobranch and liver, and affects erythrocytes possibly by the activity of PLA2 with 596U/mg of crude extract. Venom protein patterns are heterogeneous by the ability of these proteins to aggregate, the range of pI of these proteins is within a pH range of 3-7.5, mass spectrometry analysis showed that the most abundant proteins in this venom are neurotoxins with a molecular weight of 20 kDa and a metalloproteinase with a conserved sequence binding domain to Zn+2, causing bleeding observed in the main effects of this jellyfish poisoning. This work contributed to biological and toxins characterization of the jellyfish Chrysaora sp., in addition to the genetic identity of the species; a metalloproteinase was also found, characterized by a conserved novel sequence not reported to date for this jellyfish and related to cytotoxicity of the venom, causing bleeding as other metalloproteinase that found in snake venoms.

This work contributed to biological and toxins characterization of the jellyfish Chrysaora sp., in addition to the genetic identity of the species; a metalloproteinase was also found, characterized by a conserved novel sequence not reported to date for this jellyfish and related to cytotoxicity of the venom, causing bleeding as other metalloproteinase that found in snake venoms.El Filo Cnidaria contiene alrededor de 11,000 especies existentes que incluyen a las medusas, cuya característica importante es la presencia de unas células especializadas llamadas cnidocitos; las toxinas obtenidas del veneno de los cnidocitos tienen el potencial de ser una herramienta con la cual estudiar algunos sistemas fisiológicos complejos, además de poder ser utilizadas para la fabricación de nuevos fármacos y el conocimiento de su funcionamiento ayuda a la protección de posteriores envenenamientos en seres humanos; el objetivo principal de este trabajo fue evaluar y caracterizar toxinas del veneno de la medusa Chrysaora sp. colectada en la laguna Mandinga, Veracruz, México; para lograr este objetivo fue necesario el registro de parámetros fisicoquímicos, así como el estudio de aspectos biológicos, morfológicos e histológicos obtenidos en campo y en el laboratorio; algunas medusas colectadas fueron trasladadas al laboratorio y colocadas en un acuario para su mantenimiento, y otras fueron procesadas en campo para la extracción de veneno. La especie fue determinada mediante el análisis del gen de la subunidad mitocondrial citocromo c oxidasa III (COIII), y la toxicidad fue evaluada en ensayos in vivo, utilizando al pez Danio rerio como modelo biológico, y ensayos in vitro midiendo la actividad hemolítica y de fosfolipasa A2. Las proteínas presentes en el veneno se analizaron mediante geles de electroforesis de 1 y 2 dimensiones, la obtención de la huella peptídica y secuenciación de péptidos trípticos mediante secuenciación De Novo. Las condiciones adecuadas para el acuario fueron un volumen mayor a 545 L, una salinidad de 28 ‰ y una temperatura de 30 ºC. La principal afectación observada durante la aclimatación fue la eversión de la campana. La medusa presente en Mandinga es Chrysaora sp.; durante la colecta se reconocieron seis patrones de coloración que podrían estar relacionados con la salinidad; el tamaño de la medusa varia de acuerdo con la época de colecta, mostrando un mayor tamaño durante la época de lluvias (8.962.4 cm) y una predominancia de hembras con gónadas en estado maduro. El veneno causa sobre el pez Danio rerio convulsiones, parálisis, hemorragia en los ojos y disturbios en la circulación, afecta principalmente el tejido branquial, causa hiperemia en la pseudobranquia y en el hígado, y afecta los eritrocitos posiblemente por la actividad de una PLA2 con 596 U/mg del extracto crudo. Los patrones proteicos del veneno son heterogéneos por la habilidad de estas proteínas a agregarse, el rango de pI de estas proteínas se encuentra en un pH de 3- 7.5, el análisis por espectrometría de masas mostró que la proteínas que se encuentran en mayor abundancia en este veneno son neurotoxinas de peso molecular de 20 kDa y una metaloproteinasa con una secuencia de dominio conservado de unión a Zn+2 que provoca la hemorragia observada en los principales efectos del envenenamiento de esta medusa.
