| Sumario: | Mesquite (Prosopis sp.) is a legume widely used in the feeding of various species of livestock due to its high content of protein, carbohydrates, fiber, vitamins and minerals. The mesquite pod contains the phytoestrogens mesquitol, luteolin, quercetin, isoharmnetin, genistein and daidzein. It has been demonstrated that phytoestrogens can mimic the action of estrogens as they can bind to estrogen receptors α and β, so these substances could have estrogenic effects in animals and people who consume the mesquite pod for prolonged periods of time. To date, there are no studies about the possible effects that this pod could have on the reproduction and fertility of animals and humans. So, the objective of this study was to evaluate the alterations caused by the prenatal administration of the extract of mesquite pod in some reproductive aspects in the adult male and female offspring, comparing their effects with those of the phytoestrogen daidzein (DAI). Three treatments were administered for 30 consecutive days in males and females: 1) vehicle; 2) mesquite pod extract and 3) daidzein. Experimental females copulated with experimental males, and the treatments continued being administered in pregnant females throughout the gestation until offspring birth. The offspring were separated into 9 groups (n=10 each), according to the treatment administered to the parents and several reproductive variables were assessed in adulthood. In female offspring, the progression of the estrous cycle sexual behavior and serum concentrations of estradiol and progesterone were evaluated. In male offspring, sexual behavior, sperm quality and testosterone levels were assessed. The results indicated that mesquite pod extract increased the estrous cycles duration by increasing the number of days in estrus in female descendants. The percentage of receptive females, lordotic quotient, and intensity of lordosis also decreased. In male descendants, mesquite pod extract caused greater latencies of ejaculation and lower number of ejaculations, decreased glandular and testicular weights, decreases mobility, viability and sperm concentration. No significant differences were observed in female and sexual hormones in the progeny. All these effects were similar to those of DAI.
