Investigating the role of Gli2 in mouse embryonic stem cells and mouse embryogenesis

Embryonic stem cells (ESCs) are derived from the inner cell mass of the pre-implantation embryo and can recapitulate early developmental stages of the embryo. ESCs have the ability to continuously propagate while maintaining the capacity to differentiate into all cell lineages of the epiblast. The W...

ESCs have the ability to continuously propagate while maintaining the capacity to differentiate into all cell lineages of the epiblast. The Wnt/ β-catenin signalling pathway plays a fundamental role in controlling the mouse ESC regulatory circuit. However, the downstream targets of the Wnt pathway contributing to the process of pluripotency remain unknown. Here we used computational analysis to identify Gli2 as a putative downstream target of Wnt, a regulator of mESC pluripotency and a potential regulator of early embryo development. Investigating the role of this factor gave us further insight into ESC and embryo biology.
