| Sumario: | Introduction. Is estimated that 9% of individuals between 14 and 19 years has made a suicide attempt (SA), of which 61% have major depressive disorder (MDD) and often aggressive. The SA involves genetic risk factors, among which serotonergic pathway genes (SLC6A4, 5HT1Dβ, TPH2 and MAOA) and dopaminergic pathway genes (DRD2 and COMT). The aim of this study was to determine the association between polymorphisms in the afore mentioned SA adolescents with and without MDD genes. Methodology. Sample: Mexican patients diagnosed with MDD with a history of SA and MDD patients without a history SA. Genetic analysis: Genotyping by Real Time PCR with TaqMan probes for polymorphisms of genes SLC6A4, 5-HT1Dβ, TPH2, COMT and DRD2 and endpoint PCR to SLC6A4 and MAOA. Statistical Analysis: χ2 test, Fisher test and for epistasis analysis using the program MDR. Significance level p <0.05. Results. Genetic association between COMT gene and SA in Mexican adolescents with MDD was observed. In addition, it was observed that patients carrying the genotype GT of rs4570625/TPH2 had greater aggressiveness of SA. Finally, an epistatic effect between MAOA-COMT and MAOA-DRD2-DRD2 and SLC6A4 genes in Mexican SA adolescents with MDD was found. Conclusion. It is suggested that the COMT gene has an important effect on the development of the SA. Our results support the hypothesis that the effect of a gene in combination with other may be acting in synergy for the development of complex phenotypes such as SA. |
