Resumo:Active caspases 3- and -7 are considered key in somatic and testicular germ cell apoptosis signalization. Strategies of in situ or ex situ label permits to identify cell expressing active caspases. The goal of the present work was to investigate whether exists differences between percentages of sperm labeled with distinct strategies and treated with inducers or inhibitors of active caspases. Sperm samples were obtained from seven males (CII-ZV), 90 days old, sexually mature and probed fertility. Sperm with active caspases -3 and -7 were treated with the inducer imiquimod (IMQ) or inhibitor zDEVD, or both, and labeled in situ (FLICA) or ex situ (NucView), or FLICA+NucView, and evaluated by flow cytometry. No differences were observed among percentages of sperm labeled with FLICA, NuCView, IMQ+FLICA e IMQ+NucView; however differences among treatments IMQ, zDEVD or IMQ+zDEVD labeled with FLICA+NucVIew. Sperm labeled with FLICA or NucView were identified by fluorescence microscopy, but when FLICA+NucView labeling was realized sperm FLICA negatives and NucView positives were observed. There were not differences between percentages of sperm positives to FLICA or NucView; although percentages of sperm expressing active caspases -3 and -7 were observed when labeling FLICA + NucView was realized.
