description This thesis describes a novel column format to be used in pressure-driven reversed-phase liquid chromatography. The column contains an ordered pillar array where the stationary phase is supported, and it is fabricated in cyclo olefin polymer (COP) by means of standard replication techniques. The pillar array column format has shown much better efficiencies than the other traditional liquid chromatography column formats due to the order achieved with the controlled pillar packing. A particularity of this work is that the unmodified surface of the pillars acts directly as the stationary phase due to the high hydrophobicity of the COP.<br/><br/>To evaluate the performance of the fabricated column, two different experimental studies concerning the efficiency and the retention properties have been carried out using columns with the same pillar structure but with three different depths. In the first one, the influence of the depth on the band broadening effect along with the extra contribution arising from the top and bottom cover plates has been studied by injection and fluorescence detection of coumarins. In the second study, separations of different coumarin mixtures using varying mobile phase compositions have been monitored to study i) the relation between the retention factor and the ratio of organic solvent in the aqueous mobile phase and, ii) the linear relation between the retention and the surface/volume ratio, which are both predicted in the literature. The results of both studies have been compared to the case of the pillar array column fabricated in non-porous silicon showing even better results for both cases which indicates the potential this polymer column as an alternative to the pillar array silicon columns. <br/><br/>This thesis also describes the fabrication process of gold microelectrodes on the surface of the COP. In addition, the microelectrodes have been integrated into a microchannel fabricated also in COP in order to validate its response. In particular, voltammetric measurements have been performed in aqueous and organic (for the first time) media. The obtained results have been compared with the theory showing an almost perfect agreement, which open the door for the possibility to use a microfluidic system entirely made from COP with integrated microband electrodes in organic electroanalysis and in electrosynthesis.<br/><br/>Finally, a first step in order to replace the complex optical detection system used in the pillar array column characterization by a more simple electrochemical detection system is described. For that, first results on the integration of the fabricated gold microelectrodes at the end of the COP pillar array column are given at the end of this thesis.<br/><br/>KEY WORDS: On-Chip Liquid Chromatography, Cyclo Olefin Polymer, Pillar array column, Gold microelectrodes

La columna està fabricada per mètodes litogràfics de replicació en calent i conté una matriu de pilars ordenats per subjectar la fase estacionària, que simplement consisteix en la mateixa superfície no modificada dels pilars. Això és possible degut a l'alta hidrofobicitat del polímer emprat per a la fabricació del dispositiu.<br/><br/>La injecció i detecció per fluorescència de cumarines s'ha fet servir, en una primera instància, per analitzar l'eficiència de la columna. En segon lloc, l'anàlisi de les propietats retentives de la columna s'ha realitzat mitjançant l'estudi de les separacions de barreges de diferents tipus de cumarines.<br/><br/>En aquest treball també es descriu el desenvolupament i optimització del procés per a la fabricació de microelèctrodes d'or sobre la superfície del COP.
