Sumario:The study aimed to determine the degree of interaction by isobolographic analysis of co-administration in an OGD/R cell model using human SH-SY5Y neuroblastoma cells. To improve the therapeutic effect of both drugs when administered in combination. Highlights: 1. The glucose and oxygen deprivation paradigm with reoxygenation was established in the cell line SH-SY5Y. 2. Cells were tested for dapsone and cannabidiol cytotoxicity and neuroprotection concentrations. 3. Isobolographic analysis showed that dapsone and cannabidiol synergistically protect the SH-SY5Y cells. 4. In an OGD/R model in SH-SY5Y cells, DDS-CBD reduced LDH, ROS, and caspase 3. Instructions: All experiments were performed in triplicate, each repeated at least three times. In all cases, an exploratory data analysis was performed to determine whether or not there was a normal distribution, applying the Kolmogorov-Smirnov test and homogeneity of standard error, applying the Levene test. Once this was determined, parametric statistical tests such as one-way ANOVA, followed by Dunnett post hoc test and mortality in SH-SY5Y cells analysis, LC50 and EC50 values (with 95% confidence limits) were determined by Probit regression analyses. Data was analyzed using SPSS, version 25.0 software. The results of this study were performed in triplicate, with three repetitions, and were expressed as means ± SEM with A significance level of *p < 0.05, **p < 0.01, ***p < 0.001.
