CRH-expressing neurons in the medial prefrontal cortex promote social submission

The present thesis investigates the role of corticotropin-releasing hormone (CRH) release in regulating social interactions and social hierarchy in mice. Specifically, we investigated the function of medial prefrontal cortex (mPFC) neurons expressing CRH. Through a combination of electrophysiologica...

Sistema nerviosoThe present thesis investigates the role of corticotropin-releasing hormone (CRH) release in regulating social interactions and social hierarchy in mice. Specifically, we investigated the function of medial prefrontal cortex (mPFC) neurons expressing CRH. Through a combination of electrophysiological recordings, chemogenetic and optogenetic manipulations, calcium imaging, and gene silencing techniques, we demonstrate that CRH+ neurons in the infralimbic area (ILA) of the mPFC project to the rostral lateral septum (rLS) and that liberation of CRH suppresses social interactions with familiar mice. This neural circuit is therefore crucial for the familiarization process and underlies the social preference for novelty observed in mice, successfully modulating interactions between conspecifics. Motivated by these findings, I investigated the function of these cells of the mPFC for social hierarchy and showed that mPFCCRH neurons induce retreat during the tube test and therefore facilitate social submission (Graphical abstract (I).
