| Sumario: | [EN] Astrocytomas are one of the most aggressive and devastating tumors and, despite increased knowledge at the genetic and molecular level of these tumors and the development of new therapeutic strategies , the prognosis of patients with astrocytomas is still unfavorable . In the case of low-grade astrocytomas median survival is 5-10 years, whereas anaplastic astrocytomas , in general, is less than 2 years, and glioblastomas filing a survival of about one year after diagnosis . The main objective of this study was to deepen the understanding of the biology of astrocytomas and try to identify new factors that help to predict its development and evolution. So we have characterized different alterations involved in the development and progression of astrocytomas routes present in the TP53 , and IDH1/IDH2 EGFR/PI3K/PTEN genes involved in proliferation, survival and cellular metabolism , we evaluated different polymorphic variants in genes involved in the repair of DNA damage as possible risk factors for developing astrocytomas , we studied the molecular alterations of the system mismatch repair involved in the development of sporadic astrocytomas and its possible relationship with Lynch syndrome , have analyzed the role of epigenetic alterations such as methylation of promoter regions and histone deacetylase expression in the development of astrocytomas and finally , we determined the clinical and genetic variables that may be involved in the prognosis of patients with astrocytomas by developing a forecast that predicts easily individualized prognosis of patients with astrocytoma index. |
