Based on the similarity of the DNA binding domain, these proteins are grouped into different families. In plants, the MYB family of transcription factors is one of the largest and its members have been linked to different biological and biochemical processes, such as the control of cell morphogenesis, including the development of trichomes, roots and petals. Members of this family of transcription factors also act in the control of metabolism, particularly in regulating the synthesis of phenylpropanoids such as flavonoids, anthocyanins and lignins. Some members are also involved in regulating the response to phytohormones, including abscisic acid and gibberlins and also in response to environmental signals like light and availability of water. Some members of the R2R3-MYB subfamily has the ability to bind to AC elements that are commonly present in the promoters of genes participating in lignin biosynthesis, regulating specific steps of the biosynthesis of these molecules. As part of the data mining efforts of the vast amount of information available from Eucalyptus genome projects, this work performed the preliminary identification and annotation of transcription factors of the MYB family in the genome of Eucalyptus species in a attempt to identify those genes whose products are linked to lignin synthesis. Orthologous relationships between myb genes of Eucalyptus and those of other species described in the literature were inferred using phylogenetic analyses tools, allowing the identification of its putative functions. The evaluation of the expression profiles of 21,442 unigenes developed under the Genolyptus project allowed the investigation of the expression patterns of myb genes in two Eucalyptus species (E. grandis and E. globulus) and plant tissues (leaf and xylem). We also tried to integrate the information provided by published genetic maps of microsatellite markers containing QTLs for lignification with data from the Eucalyptus genome sequencing project. Co-localization of myb genes with QTLs for these characters has allowed the identification of myb genes of interest for future studies in functional genomics. We identified 212 genes belonging to the MYB family of transcription factors in the genome of different species of Eucalyptus. Each of these genes was submitted a process of manual annotation, and 177 of these were classified as myb-R2R3, and characterized in terms of its ultra-structure of introns and exons. The analysis of the results obtained by the use of three different methodologies: molecular phylogeny, analyses of expression and co-localization with QTLs, allowed the identification of candidate genes that are likely involved in the control of lignin content and quality in Eucalyptus. EucMYB_024, EucMYB_032, EucMYB_044, EucMYB_046, EucMYB_050 and EucMYB_068 showed evidence under the three methodologies that suggest they are involved in an important way in the process of xylogenesis in Eucalyptus. The results herein are useful in allowing the identification of target genes for future studies in functional genomics. Apparent functions were associated with these six genes. The understanding of their role in the development of Eucalyptus wood will be effectively established in future studies of genetic transformation, where the effects of overexpression and silencing of these genes may be assessed.

Com base na similaridade de seus domínios de ligação ao DNA, estas proteínas são agrupadas em diferentes famílias. Em plantas, a família MYB de fatores de transcrição é uma das mais numerosas e seus membros têm sido relacionados a diferentes processos biológicos e bioquímicos, como por exemplo o controle da morfogênese celular, incluindo o desenvolvimento de tricomas, raízes e pétalas. Os membros desta família de fatores de transcrição também atuam no controle do metabolismo, particularmente na regulação da síntese de fenilpropanoides, como os flavonoides, antocianinas e ligninas. Alguns membros também estão envolvidos na regulação da resposta a fitormônios, incluindo ácido abcísico e giberelinas e também em resposta a sinais ambientais como a luz e a disponibilidade de água. Alguns membros da subfamília MYB-R2R3 têm a capacidade de se ligar a elementos AC que são comumente presentes nos promotores de genes que participam da biossíntese de lignina, regulando etapas específicas da biossíntese destas moléculas. Como parte dos esforços de mineração da enorme quantidade de informações disponibilizadas no âmbito de projetos de genômica em Eucalyptus, o presente trabalho se propôs a realizar a identificação e a anotação preliminar de fatores de transcrição da família MYB presentes no genoma das espécies de Eucalyptus, em uma tentativa de se identificar aqueles genes cujos produtos estão relacionados ao controle do processo de xilogênese. Pelo uso de ferramentas de filogenia molecular foram inferidas as relações de ortologia entre genes myb de Eucalyptus e aqueles de outras espécies descritos na literatura, permitindo a identificação de suas funções aparentes. Um ensaio de avaliação dos perfis de expressão de 21442 unigenes desenvolvido no âmbito do projeto Genolyptus, permitiu a investigação do padrão de expressão dos genes myb em diferentes espécies (E. grandis e E. globulus) e tecidos da planta (xilema e folha). Foi ainda realizado um esforço para integração das informações fornecidas pelos mapas genéticos de marcadores moleculares microssatélites publicados na literatura, contendo QTLs relacionados ao processo de lignificação, com os dados de sequenciamento do genoma de Eucalyptus. A co-localização dos genes myb com QTLs para estes caracteres permitiu a identificação dos genes myb de maior interesse para a realização de estudos futuros de genômica funcional. No presente trabalho foram identificados 212 genes pertencentes à família de fatores de transcrição MYB no genoma de diferentes espécies de Eucalyptus. Cada um destes genes foi submetido a um processo de anotação manual, sendo que 177 destes foram classificados como myb-R2R3, e caracterizados quanto à sua ultra-estrutura em termos de íntrons e éxons. A partir da análise dos resultados obtidos pelo uso de três metodologias distintas: reconstrução filogenética, análise de expressão e co-localização com QTLs, os genes candidatos a exercerem participação efetiva no controle do teor e qualidade da lignina em Eucalyptus foram sugeridos. Os genes EucMYB_024, EucMYB_032, EucMYB_044, EucMYB_046, EucMYB_050 e EucMYB_068 apresentam evidências, segundo as três metodologias utilizadas, que sugerem que estejam participando de maneira importante no processo de xilogênese em Eucalyptus. Resultados desta natureza são úteis no sentido de permitirem a identificação de genes alvo para realização de estudos de análise funcional. Neste trabalho, funções aparentes foram associadas a estes genes.
