Sumario:It is known that about 150 thousand years ago man already used medicinal plants to treat ills and health problems. The “in natura” consumption of plants or even in the form of derived products is directly linked to adverse reactions, justified by their increasing use. Among all the species found is Rhamnus purshiana, also popularly known as Cascara Sagrada, being used due to its laxative effect. However, there are still few studies related to its use. The present work aimed, through the crude extract of the barks of Rhamnus purshiana, to carry out the in silico analysis of the major compounds mentioned in the literature, and to predict the oral bioavailability, the toxicological potential and the pharmacokinetic data of Cascara Sagrada using the Molinspiration, Protox software. II and AdmetSAR. In addition to these, perform the quantification of secondary metabolites and the antioxidant capacity by the DPPH radical capture method and microbiological through the disk diffusion assay and minimum inhibitory concentration. The Rhamnus purshiana extract after the analysis showed good oral bioavailability, in addition to immunotoxic probability and mutagenicity. Excretion, on the other hand, showed good elimination, when the compound is already biotransformed. As for the determination of polyphenols and flavonoids, it showed concentrations of 4.00 ± 0.27 mg/g and 2.98 ± 0.11 mg/g of extract, which were determined by phytochemical dosages, respectively. Rhamnus purshiana was able to capture reactive oxygen and nitrogen species with an antioxidant capacity of IC50=1.78 ± 0.10 mg/ml. Antimicrobial activity was not significant against Pseudomonas aeruginosa, Escherichia coli, Serratia marcences Staphylococcus aureus, Streptococcus mutans, Enterococcus faecalis, Candida albicans. In this way, through the positive results, it can be suggested the use of Rhamnus purshiana in an effective and safe way.
